提出一新觀點,指出果蠅軸突導(dǎo)向中確有信號分子受體的作用存在。
人類唐氏綜合癥細(xì)胞粘連分子在果蠅中的同源物是Dacam,它與Dock相互作用,進(jìn)而得到分離,Dscam是免疫球蛋白超家族中的一員,其結(jié)構(gòu)與許多常見的軸突導(dǎo)向受體相似。Schmucker etal.發(fā)現(xiàn),Dscam在軸突中表達(dá),其突變體會引起導(dǎo)向出現(xiàn)缺陷,而Dscam的基因結(jié)構(gòu)更是讓人大吃一驚,Dscam基因具有相當(dāng)多的剪切方式,形成蛋白千變?nèi)f化,但都有相同的結(jié)構(gòu)域,如果每種剪切方式都產(chǎn)生蛋白,那么將有38,000種之多,在神經(jīng)系統(tǒng)尚未發(fā)現(xiàn)這種好象免疫抗體類蛋白才會有的現(xiàn)象。
Dscam好象是探測器的接收器,那么誰是派出的探測器呢?Dscam的配體還未確定。它的人類同源物也是軸突導(dǎo)向受體嗎?
