銀杏內(nèi)酯B對腦缺血-再灌注神經(jīng)元損傷的保護作用

【摘要】 目的:研究銀杏內(nèi)酯B對大鼠腦缺血-再灌注神經(jīng)元損傷的干預(yù)作用及其可能機制。方法:采用Kameyama的3條動脈夾閉法制備大鼠腦缺血-再灌注損傷模型。45只Wistar大鼠隨機分成假手術(shù)對照組、缺血-再灌注模型組、生理鹽水對照組和銀杏內(nèi)酯B預(yù)處理組;測定各組動物腦勻漿液中超氧化物歧化酶(SOD)、谷胱甘肽過氧化物酶(GSH-Px)和ATP酶活性以及丙二醛(MDA)含量,并觀察腦組織的病理學(xué)改變和細胞凋亡指數(shù)。結(jié)果:全腦缺血-再灌注可引起SOD、GSH-Px和ATP酶活性明顯下降,MDA含量顯著升高;腦組織呈現(xiàn)炎癥性改變,層次不清,細胞凋亡指數(shù)達(40.2±6.3)%。銀杏內(nèi)酯B(1~10 mg/kg)能劑量依賴性地降低MDA水平,提高SOD、GSH-Px和ATP酶活性(P<0.05或P<0.01),明顯減輕腦組織神經(jīng)細胞損害,減少細胞凋亡發(fā)生率。結(jié)論:銀杏內(nèi)酯B對大鼠腦缺血-再灌注神經(jīng)元損傷具有明顯的保護作用,其機制可能是通過減少自由基的產(chǎn)生和增強ATP酶活性。

【Abstract】 Objective: To investigate the protective effects and mechanism of ginkgolide B(銀杏內(nèi)酯B, GB) on neuron injury induced by cerebral ischemia/reperfusion(I/R) in rats. Methods: The bilateral cerebral hemisphere I/R model was established by ligating three arteries with Kameyama's manner. Forty-five Wistar rats were randomly assigned to sham operation control, I/R model, normal saline control (NS) and GB pretreatment group. The activity of superoxide dismutase(SOD), glutathione peroxidase(GSH-Px), ATPase, and the contents of malondialdehyde(MDA) in brain tissue were measured.
