[摘要] 目的 檢測鼻咽癌CNE1細(xì)胞X射線照射前后多藥耐藥基因(mdr1基因)及其編碼產(chǎn)物P糖蛋白(P-glycoprotein,P-gp)是否表達,以及化療藥物敏感性的變化。方法 利用RT-PCR、Western blotting和流式細(xì)胞儀(FCM)檢測CNE1細(xì)胞X射線照射前后的mdr1基因和P-gp的表達及對柔紅霉素(daunorubicin,DNR)的外排功能。結(jié)果 鼻咽癌CNE1細(xì)胞射線照射前mdr1基因、P-gp不表達;射線照射后較長時間內(nèi)mdr1基因、P-gp均長時間表達。CNE1細(xì)胞射線照射后對DNR的蓄積作用略有降低。結(jié)論 鼻咽癌CNE1細(xì)胞X射線照射后化療敏感性降低。[關(guān)鍵詞] 鼻咽腫瘤;腫瘤細(xì)胞,培養(yǎng)的;基因,mdr1;P-糖蛋白;放射治療;化療敏感性
The change of chemotherapy sensitivity of nasopharyngeal carcinoma cell after radiotherapy

[Abstract] Objective To examine the expression of multidrug resistance gene(mdr1) and P-glycoprotein(P-gp) in nasopharyngeal carcinoma(NPC) CNE1 cell before and after radiotherapy,as well as the changes of chemotherapy sensitivity.Methods The expression of mdr1 gene and its protein P-gp was assayed by reverse transcription-polymerase chain reaction (RT-PCR) and Western blotting.Flow cytometry (FCM) was performed to examine the function of cell efflux to daunorubicin.Results Expression of mdr1-mRNA and P-gp of CNE1 cell in long time after radiotherapy was found,but not before.Intracellular daunorubicin(DNR) accumulation of CNE1 cell decreased after radiotherapy.Conclusion Irradiation decreases the chemotherapy sensitivity of CNE1 cell. ......
